A Comparison of the TC10 Automated Cell Counter and the Hemocytometer for Quantifying Insect Hemocytes

چکیده
Introduction Hemocytes are an essential component of the invertebrate innate immune system (Strand 2008). Their close similarities to vertebrate white blood cells enable their use as a tractable model for immunological research. The quantification of hemocytes in the hemolymph (invertebrate blood) is one way of assessing the efficacy of the immune response because the absolute number of hemocytes changes during the course of an infection (Bergin et al. 2003, Brayner et al. 2007), and the density of hemocytes in the hemolymph is strongly correlated with the capacity to encapsulate or phagocytose parasites (Prevost and Eslin 1998, Kraaijeveld et al. 2001, Wilson et al. 2003, Costa et al. 2005). Most researchers continue to use cell staining and manual microscopy techniques to enumerate the hemocytes in a given sample. However, given the laborious nature of this method (with a throughput often as low as 6–8 samples/hr), more rapid techniques are required to increase our understanding of the role of hemocytes in invertebrate immunology.

Insect hemocytes have been the focus of researchers for decades (e.g., Jones, 1962; Ravindranath, 1978; Hall, 1983; Ribeiro & Brehelin, 2006). Most research has sought to increase our understanding of what factors influence hemocytes numbers, especially in response to various stressors (e.g., Silva et al., 2002; Andrade et al., 2003; Mochiah et al., 2003). Moreover, the recent increase in quest...

The aim of the present study was to determine various types of hemocytes, total and differential hemocyte count and to show some indices of phagocytosis, including percent phagocytosis and phagocytic index in Indian white shrimp, Fenneropenaeus indicus in Iranian shrimp farms. The hemolymph was extracted from the shrimps and mixed with anticoagulant. It then stained and Yeast was added as forei...

متن کاملConstruction of a Recombinant Bacmid DNA to Express Influenza Virus Matrix Protein1 (M1) in Insect Cell Line
Background and Aims: Virus-like particles (VLPs) have been suggested to be a promising recombinant vaccine approach. Several studies have reported that the influenza VLPs produced in insect cells is an effective vaccine candidate. Due to crucial role of matrix M1 protein in assembly and budding of Influenza particles, in all VLPs structures, M1 protein have been considered as a main component. ...
متن کاملValidity of Selected WBC Differentiation Flags in Sysmex XT-1800i
Background: Automatic Cell Counter devises make the CBC differential very easy and delivering the results in few second. However, the problem with this device is facing a flag requires a time-consuming microscopic review of the specimen which causes unacceptable wait times for patient as well as costs for laboratories. In this study, we calculated the validity of WBC d...
